Our Core Facility Metabolomics operates across three laboratories equipped with state-of-the-art instrumentation for targeted, untargeted, quantitative, as well as spatial metabolomics and lipidomics. Equipment

At our Metabolomics Core Facility, we offer specialized sample preparation techniques, including both one-phase and two-phase extractions tailored to different biological matrices and metabolic profiles of interest. To enhance metabolic recovery, Focused Ultrasonication™ is employed, ensuring efficient and reproducible extraction. We process a wide range of sample types, including solid tissues such as stool, brain, and liver, as well as liquid samples like urine, blood serum, cerebrospinal fluid (CSF), and coculture media.
For MALDI imaging, we meticulously prepare cryosectioned samples by applying matrix solutions using a precision sprayer. The choice of matrix depends on the metabolites of interest, optimizing sensitivity and resolution for high-quality spatial metabolomic analysis.

LE220-plus Focused-ultrasonicator

The LE220-plus Focused-ultrasonicator is a scalable, automated and batch-reproducible ultrasound sample preparation instrument. Designed for integration with high-throughput laboratory automation, the LE220-plus includes an integrated AFA ultraCUBE water chiller and a water conditioning module and is used for the extraction of polar metabolites and lipids from solid and semi-solid sample.

LEICA DM 1000 LED

In the same pipeline we use the LEICA DM 1000 LED to assess spraying performance in the quality control workflow for MALDI imaging. This highly versatile and high-quality microscope is ideal for clinical and laboratory use.

We employ state-of the-art mass spectrometry to produce high quality data. The principle is based on the formation of charged ions from the analyte molecules.
Compared to classic detection methods such as UV/Vis or RID, mass spectrometry also contains information about the molecular mass (m/z values) and the fragmentation (MS/MS) of the analyzed compounds. In untargeted analyses, this provides useful criteria for identifying compounds, and gain first structural information about unknowns. For targeted approaches, this technique allows for method optimization based on specific structural properties of the analyte, greatly improving specificity and selectivity.
